Tissue engineering is gaining interest as it is applied for regeneration of organs to attain their lost function. BMPs have been successfully used to regenerate the bone in humans. Most recent preclinical data suggest that BMPs have a potential to provide protection against inflammation and fibrosis in acute and chronic injury of parenchymal tissues when applied systemically to sustain the function of kidney and liver. The application of BMPs from a local to systemic utility is a rapidly growing field, gaining interest among researchers and biotech entrepreneurs. In this volume, we summarize the advances made on thelocal and systemic use of BMPs including chapters covering the regulation of BMP-signalling pathways, biological actions of BMPs in bone, cartilage and teeth, as well as clinical applications and potential systemic use of BMPs for tissues beyond bone. Completes the series of PIR-volumes on BMPs by S. Vukicevic/K.T. Sampath with most recent data (Bone Morphogenetic Proteins - Regeneration of Bone and Beyond / 2004; and Bone Morphogenetic Proteins - From Laboratoryto Clinical Practice / 2002) INDICE: From the contents: An update on BMP biology.- Regulation of BMP-2 and GDF5 signaling by ligand mutants and receptor endocytosis.- Signaling crosstalk.- Generation of conditional alleles for BMP-2 and BMP-7.- BMP-2 and BMP-4 action in bone.- Role of BMP-4 in tooth differentiation and bone morphogenesis.- Alterations of BMP signaling pathway(s) in skeletal malformations.- BMP antagonists and kidney.- SOST related biology.- BMP-7 induced tumor-stroma interactions in bone metastasis.- Endocultivation of vascularized bone segments.- Induction of cementogenesis and periodontal tissue regeneration by the bone morphogenetic proteins.- BMP and cartilage biology.- BMP-1 and its isoforms in tissue regeneration.- Clinical use of rhBMP-2 in the spine.- Use of BMP-2 in acute orthopedic trauma.- Reconstruction of maxillary and mandibular defects using BMP-2.- New carriers of BMPs.- Systemic administration of BMPs.
